The most honest summary of the evidence is this: melatonin can help shift the timing of sleep (jet lag, shift work, delayed body clock) and offers only a modest nudge for ordinary insomnia, while magnesium, glycine, L-theanine and valerian have weaker, more mixed evidence. For persistent insomnia, structured behavioral treatment and better sleep habits outperform almost every pill on the shelf.

Before any supplement: what works better

It is worth being blunt, because the supplement aisle rarely is. For chronic insomnia, the first-line treatment recommended by major bodies is not a pill at all. Cognitive behavioral therapy for insomnia (CBT-I) — a short, structured program addressing sleep timing, unhelpful thoughts and time spent awake in bed — is recommended as first-line care by the American Academy of Sleep Medicine and others, and its benefits tend to last longer than medication.

Alongside it, the unglamorous basics do more than most capsules: a consistent wake time, a cool and dark room, limiting alcohol and caffeine later in the day, morning daylight, and a wind-down routine. The NHS and CDC both publish plain-language sleep-hygiene guidance. If you have tried these and still struggle for weeks, that is a reason to see a clinician — not to buy a stronger supplement.

1. Melatonin — the one with real, if narrow, uses

Melatonin is a hormone your body makes as darkness falls; it signals timing, not sedation. That distinction matters. Its strongest role is resetting a misaligned body clock — easing jet lag, helping shift workers, or nudging a delayed sleep phase earlier. For ordinary trouble falling asleep, reviews suggest it can shorten the time to fall asleep somewhat, but the effect is generally modest. Mayo Clinic and the NHS both describe it as most useful for timing-related sleep problems.

Practical points: timing and low doses tend to matter more than large ones — taking a small amount earlier in the evening is often more effective for shifting the clock than a big dose at bedtime. In the United States melatonin is sold as a supplement, so it is not tightly regulated, and independent testing has repeatedly found that the actual content can differ substantially from the label. Buy from a reputable brand and treat "more" as a red flag, not a feature.

Safety: melatonin can cause next-day grogginess, headache or vivid dreams, and it can interact with several medications — including blood thinners, some blood-pressure and diabetes drugs, immunosuppressants, sedatives and certain antidepressants. If you are pregnant or breastfeeding, or take any of these, ask a clinician before starting. It is not a "harmless natural" default at high doses.

Magnesium is having a moment, and it is an essential mineral involved in nerve and muscle function. But the leap from "essential mineral" to "sleep aid" is not well supported. The clinical trials are small, short and inconsistent, and there is no strong evidence that supplementing improves sleep in people who are not deficient. The NIH Office of Dietary Supplements notes magnesium's roles but does not endorse it as a proven treatment for insomnia.

If your diet is genuinely low in magnesium, correcting that is reasonable for general health, and dietary sources — nuts, seeds, legumes, leafy greens, whole grains — are the sensible first stop. High-dose supplements commonly cause loose stools or cramping, and people with kidney disease should not supplement without medical advice, because they cannot clear excess magnesium normally.

3. Glycine — early promise, little proof

Glycine is an amino acid, and a handful of small studies have hinted that a few grams before bed might help people feel less unrefreshed the next day, possibly by slightly lowering core body temperature. The signal is interesting but preliminary — the studies are few and small, and there is little data on long-term use. It is reasonable to file glycine under "low-risk, unproven" rather than recommend it with confidence.

4. L-theanine — calm, not sleep

L-theanine, an amino acid found in tea, is often marketed for sleep but is better understood as a mild relaxation aid than a sedative. Evidence that it meaningfully improves objective sleep is weak. It appears low-risk for most adults, but "unlikely to hurt" is not the same as "shown to help," and it should not displace addressing the actual cause of poor sleep.

5. Valerian — the classic herb with muddy data

Valerian root is one of the oldest herbal sleep remedies, yet decades of research remain inconsistent — some trials show a small benefit, many show none, and the products themselves vary widely in what they contain. The National Center for Complementary and Integrative Health summarizes the evidence as limited and mixed. Valerian can cause drowsiness and, rarely, has been associated with liver concerns; combining it with alcohol or sedatives is not advised.

Sleep in menopause: a special case

Sleep disruption is one of the most common and under-discussed symptoms of the menopause transition. Night-time hot flashes and sweats fragment sleep, and mood changes and anxiety compound it. The uncomfortable truth is that supplements are rarely the answer here, because the driver is often the vasomotor symptoms themselves. The Menopause Society and ACOG discuss evidence-based options — which may include treating hot flashes directly and behavioral strategies for sleep — that are more likely to help than a magnesium capsule.

Melatonin's timing effects may be relevant if your sleep schedule has drifted, but it will not touch a hot flash that wakes you at 3 a.m. If night sweats or early waking are wrecking your sleep, that is a conversation to have with a clinician about the whole picture, not a problem to solve alone in the supplement aisle.

How to try a supplement sensibly

  • Fix the basics first. Consistent wake time, morning light, less late alcohol and caffeine, and a dark, cool room do more than most capsules.
  • Change one thing at a time. Stacking several supplements makes it impossible to know what — if anything — is working.
  • Start low, keep it short. Especially with melatonin, small doses are often as effective, and there is little reason to escalate.
  • Check interactions. If you take prescription medication or have a chronic condition, ask a pharmacist or clinician before starting.
  • Give it a defined trial. If a few weeks bring no clear benefit, stop rather than adding more.

When to see a doctor

Please treat ongoing insomnia as a medical issue, not a shopping problem. See a clinician if poor sleep lasts more than a few weeks, disrupts your daytime functioning, or comes with loud snoring and gasping (possible sleep apnea), restless legs, or low mood and anxiety. Chronic insomnia warrants proper assessment, and CBT-I is the recommended first-line treatment.
